Alzheimer’s disease (AD) is the most well-known and most common type of age-related dementia. Amyloid deposition and hyperphosphorylation of tau protein are both pathological hallmarks of AD. Using a triple-transgenic mouse model (3xTg-AD) that develops plaques and tangles in the brain similar to human AD, we provide evidence that active full-length DNA amyloid-β peptide 1–42 (Aβ42) trimer immunization leads to reduction of both amyloid and tau aggregation and accumulation.

Methods
Immune responses were monitored by enzyme-linked immunosorbent assay (ELISA) (antibody production) and enzyme-linked immunospot (cellular activation, cytokine production). Brains from 20-month-old 3x Tg-AD mice that had received DNA Aβ42 immunotherapy were compared with brains from age- and gender-matched transgenic Aβ42 peptide-immunized and control mice by histology, Western blot analysis, and ELISA. Protein kinase activation and kinase levels were studied in Western blots from mouse hemibrain lysates.

Results
Quantitative ELISA showed a 40% reduction of Aβ42 peptide and a 25–50% reduction of total tau and different phosphorylated tau molecules in the DNA Aβ42 trimer-immunized 3xTg-AD mice compared with nonimmunized 3xTg-AD control animals. Plaque and Aβ peptide reductions in the brain were due to the anti-Aβ antibodies generated following the immunizations. Reductions of tau were likely due to indirect actions such as less Aβ in the brain resulting in less tau kinase activation.

Conclusions
The significance of these findings is that DNA Aβ42 trimer immunotherapy targets two major pathologies in AD—amyloid plaques and neurofibrillary tangles—in one vaccine without inducing inflammatory T-cell responses, which carry the danger of autoimmune inflammation, as found in a clinical trial using active Aβ42 peptide immunization in patients with AD (AN1792)


Bit past my bedtime, just thought people might find this interesting.

A different sort of amyloid tangle is involved in damage to beta cells in type II diabetes.

Sounding like a broken record (or an mp3 file corruption): Again, are we really focusing on root cause or just symptoms that happen to be present? We've been very successful in the past with pharmaceuticals in controlling individual symptoms at a very high cost to the individuals who are dealing with broader metabolic system issues.

It's good to use research to understand the details and associated functions of how a disease presents, but not to then use a pill to reduce associated symptoms thinking we've got the answer.

This may be useful to triple-transgenic humans. Or at least to some people with very high genetic risk of alzheimers due to amyloid plaque. This seems a bit risky, since humans are not generally triple transgenic mice, just because this isn't causing "friendly fire" more generalized autoimmune issues in the mice, I don't think that means that it won't cause it in humans.

Alzheimer's is so bloody awful, I don't really think they should leave a stone unturned.

Agreed, and since AD has been on the increase over the recent past, it's helpful to turn every stone to determine what has changed to cause this increase. The price paid by AD families is horrendously high.

As I hve been looking to PREVENT family history from repeating itself,and any doctor yet visited says "take this pill", or "surgery" or " no surgery" , the efforts to "prevent" issues of all types is still in the dark ages.

My grandfather lived to mid 90's ( died due to fall carrying in his new tv) and his younger sister lived to 105; at 100 she was rather lucid, the last time I saw her. Grandfathers first wife ( and her sisters, except one) all died of breast cancer. Second wife died of AD. All tall thin trim, well dressed women. Otherwise very healthy. GO figure.

Im no longer keen on the vaccines for the panacea, those too have problems that doctors sweep under the rug.BECAUSE THEY ARE NOT LIABLE. I often wonder now if my son is autistic because of the vaccines with thimerisol. IT is the ONE chemical I am allergic to---

As an infant, ANd I asked pediatric doc about delaying and changing the vaccination protocol, she jumped down my throat making me out to be a bad mother. Yet, in the horse world we have changed the vaccination program ( delayed) because the vaccines were interfering with the protection given by the mothers milk.

And a friend who breeds dogs, gives only the bare minimum of vaccines.

Yet we are not allowed to do that for our children. I do believe in the judicious use of vaccines. Just not the endless poking.

We are guinea pigs and lemmings.........too angry to continue politely.....

Taking a look at the CDC's Pink Book of Vaccine Ingredients was enough for me! It really is chilling to see the ingredients being injected into otherwise healthy little children, at ever increasing rates!! When my kids were young, I believe they got a total of 7 vaccines - today it is at 66 and rising by the time they are 18!! Big Pharma is lining their pockets for sure....
