Will glutamine help you avoid pain if you take it every once in a while? For example, if you know you're going to help someone move or something like that, can you take it beforehand like you would an acid reducer to avoid heartburn when you know you're going to eat something scary? Or does it have to build up in your system and be taken everyday?

It would be taken AFTER the work was done, but yes, it can be used in that way to help with recovery.

Actually, alot of the research on L-glu is from burn pateints and post-surgical patients who were supplemented with high doses (60g - 80g) per day in divided doses and their recovery times were greatly reduced.
